• Immunization technology
PX’Monoclonals will develop substractive immunization approaches
through a collaboration with the INSERM. This specific immunization
method aims at generating antibody directed against weakly antigenic
and rare epitopes and involve a distinct immune tolerization
of the host animal.

• Humanized
mice
In
a joint effort, PX’Monoclonals and the company Genoway will work
on the generation of transgenic mice capable of producing humanized
antibodies. This strategy is based on the replacement and/or the
insertion of human sequences to humanize the constant or variable
parts of antibodies. Those technologies may also be used for antibody
« animalization ».

• Humanized
Antibodies
Focusing
on Protein’eXpert competences in terms of molecular biology and
recombinant proteins production (Fab/ScFv), PX’Monoclonals will
also launch a programme adressing the development of humanized
recombinant antibodies.
This approach will be based on RT-PCR cloning of antibody variable
part, selection of the DNA sequence related to IgG constant part,
CDR grafting, potential humanization of variable fragments and
eventually transfer of the sequence in an expression vector and
transfection of a mammalian cell line (eg CHO) to produce the antibody.

• Human
Antibodies
The
development of fully human monoclonal antibodies will also be targeted
in close cooperation with the INSERM. This programme will consist
in the selection of seroconverted patients (patients resistant
to a specific infection), patient blood collection, isolation of
B lymphocytes, cloning by limited dilution, EBV-immortalization
and culture of transformed B cells to produce the human monoclonal
antibodies.
Alongside the focus on humanization of therapeutics antibodies,
the platform will also develop specific technologies related to
the antibody production process (immunization, selection of clones,
